Cabozantinib (marketed under the tradename Cometriq, formerly known as XL184) is a potent VEGFR2 inhibitor with IC50 of 0.035 nM. XL-184 also displays potent activity against Met, Ret, Kit, FLT1, FLT3, FLT4, Tie2, and AXL. XL184 at low concentration(0.1-0.5 μM) is sufficient to induce marked inhibition of constitutive and inducible Met phosphorylation and its resultant downstream signaling in MPNST cells, and inhibit HGF-induced MPNST cell migration and invasion.

How cabozantinib works

 

Cabozantinib is a cancer growth blocker. It works by blocking particular proteins called protein kinases on cancer cells. The protein kinases encourage the cancer to grow. Cabozantinib blocks a number of these proteins and is called a multi kinase inhibitor. It also stops the cancer cells growing blood vessels, which the cells need to be able to grow.

Cabozantinib may shrink the cancer or stop it growing for a time.

In B16F10 cells, cabozantinib inhibits HGF-inducedmigration and invasion with IC50 values of 31nM and 9nM, respectively. Moreover, cabozantinib shows anti-proliferation efficacy in a variety of tumors such as SNU-5, Hs746T, MDA-MB-231 and U87MG. It is also reported that the combination of cabozantinib and gefitinib can cause potent inhibition of the gefitinib-resistant HCC827GR6 cell line.
